Mathematical modeling is an important tool to support the management of water resources. This study proposes a mathematical model to simulate self-purification in watercourses. In the study, the temperature of a jar test was adjusted by a water bath in a range of 20 to 28 °C, and the water flow velocity was maintained between 0.29 and 0.87 m s-1, conditions observed in natural watercourses. The kinetic experiments, which treated synthetic wastewater with an initial chemical oxygen demand concentration of 50 mg L-1, were conducted according to a Central Composite Design. From the statistical adjustment of the chemical oxygen demand values, a mathematical equation was generated for each self-purification day. The mathematical model was validated in a self-purification test at 26 °C and 0.38 m s-1, with an initial chemical oxygen demand concentration of 30 mg L-1. The proposed model satisfactorily describes the total variation of responses, with a ratio of 99.46%.
