VITAMIN D SUPPLEMENTATION, ANTIOXIDANT ACTIVITY AND LIVER FUNCTION IN AN EXPERIMENTAL MODEL OF TYPE 2 DIABETES MELLITUS

Abstract

The study aimed to evaluate the effect of vitamin D supplementation on antioxidant activity and liver function in an experimental model of type 2 diabetes mellitus in humans. A trial was carried out with 35 male Wistar rats to induce DM2 through a high-fat, normoprotein diet followed by the administration of streptozotocin (30 mg/kg i.p.). After confirming diabetes, the rats were divided into five groups: normal control, diabetic control, diabetics treated with metformin 150 mg/kg, and diabetics supplemented with vitamin D at doses of 0.25 µg/kg/day and 0.50 µg/kg/day. The normal control group received only citrate buffer. The treatment period was 28 days. The results showed that the vitamin D-treated groups showed a significant reduction in mean fasting blood glucose levels compared to the diabetic control group. In addition, vitamin D supplementation resulted in increased activity of the antioxidant enzyme SOD and higher concentrations of GSHNP, however, there were no significant differences in catalase activity between the groups. With regard to liver function, the group that received 0.50 µg/kg/day of vitamin D showed a reduction in total hepatic cholesterol and mean TGO levels. Histopathological analysis revealed the presence of binucleated cells in the livers of rats treated with vitamin D, a possible indication of cell regeneration. Therefore, vitamin D supplementation for four weeks improved fasting blood glucose, the activity of the antioxidant enzyme SOD and liver function in an experimental model of diabetes.
